When it comes to multiple characters in the same shot I definitely would do both characters to the same level of animation at the same time (whether or not it is an action sequence - because no matter what, both characters will be interacting either directly (physically) or indirectly (emotionally)).

So I would block character A, and then character B. I would then go back and tweak A and then B until I’m happy with their interaction (physical or emotional interaction). Then I would do the rough lip sync/facial animation on A and B. Then I would go and spline and finalize the animation and lip sync for A and B, adjusting A or B as necessary (especially at the contact points, if they contact).
