how do I disable an animation layer?

There is no way to mute the layer at the moment. if you are doing this in story treating each clip as a layer you can.

the hack for mute layer in mobu right now would be to go to frame -1 and do a 0 key, now select all of the keys and move them so that 0 key is past the last frame of your animation ( this takes the weight to 0 for the entire time basically.
