I wouldn't use the clone stamp on this, because you will have to do it frame by frame.
Roto or key it out and duplicate the layer.

I've never actually used the Clone Stamp tool in After Effects. I use it in Photoshop to create my clean plates, but that's it.
As far as getting a copy of a car, the way I would do it is that I would make a copy of the layer, move it ahead a few frames so the car is in front a bit, and then do a roto around it. It would give you the same effect, but it might take more time than using the Clone Stamp Tool. But like I said, I'm not familiar with it in AE, so I would use the other technique.

It seems that you missed a point. Or one aspect. Whatever. The thing is that you have to double-click the layer you want to use your clone-tool on. The tools like paint and clone(stamp) tool are working in a way that in order to see them you have to work with layer source window. To open it - you have to, as I've said earlier, double-click the layer you are trying to edit.
