Just before the "Ready?" text appear there are some animated glowing swirling lines - how is this done? Could you do it in Combustion only or do you need other software?

I can tell you now that it could be done in combustion (as in, with no plugins) but i will bet money that that was created with Trapecode's 3Dstroke (possibly with combustion). I have some work to do in combustion tonight so if i get a chance , i'll show you what this little gem can do (rather quickly)

The moving glowing lines is trapcode 3d stroke (http://www.trapcode.com/products_3dstroke.html), not particular (http://www.trapcode.com/products_particular.html).

no, see particular, 3dstroke I think is more manual, but particlar has this aswell.

Wow i didn't know particular got down like that. While 3dstroke is more manual it was built for that purpose alone, so it has greater control over every curve.
