Does anyone give me an idea, how I can create a flexible rope?
If I create with nurbs a thin cylinder, is that the right way? And when it is, what is my next step?

I would use an EP curve with history attached as the deformation object that way you would have alot more control over the way it moves, and can add dynamics to it very easily with goal wieghts to make it look like rope.

Another way would be to use the IK spline tool, which is great for things like a rope. After browsing throught that tutorial that was posted this is a smilar way, but I think that the tutorials that ship with maya are alot better then that tutorial posted.

If you need more help with these things they are in the help files, with some great tutorials.
